Protein import through the nuclear pore complex

Nuclear transport receptor NTF2 is responsible for the import of a cargo protein through binding to RanGDP; however, I am curious how it(NTF2) leaves nucleus back to cytoplasm as I couldn’t find a definite answer on the Internet

NTF2 has a high affinity towards Ran-GDP and mediates translocation of this complex through the NPC to the nucleus. In the nucleus RanGEF releases Ran by exchanging GTP to GDP, for which NTF2 has no significant affinity. As a result, unbound NTF2 is exported back into the cytoplasm. However, I am unaware of exact mechanism for NTF2 transport from nucleus to the cytoplasm.

Here’s what I found from the 2012 paper.

“ Regulation of translocation of NTF2 to the cytoplasm could be achieved by two possible mechanisms.

The first is that the function of a nuclear protein, which is regulated, is responsible for controlling the nuclear-cytoplasmic trafficking activity of NTF2, and Tween-20 treatment may lead to inhibition or activation of the function of this protein, causing inhibition of NTF2 translocation to the cytoplasm.

The second is that interaction between NTF2 and specific nucleoporins on the nucleoplasmic side of the NPC is regulated by reversible modification of the nucleoporins, as the transport activities of receptors have been shown to be affected by phosphorylation of Nups [[52]]

I also asked qualified people and got the following answer:

NTF2 moves back and forth across the NPC rapidly, regardless of whether RanGDP is bound or not.
